Region hit by Sandy struggles to resume daily life

NEW YORK (AP) ? Two major airports reopened and the floor of the New York Stock Exchange came back to life Wednesday, but across the river in New Jersey, the National Guard searched for flood victims and fires still raged two days after Superstorm Sandy.

For the first time since the storm battered the Northeast, killing 55 people and doing billions of dollars in damage, brilliant sunshine washed over the nation's largest city ? a striking sight after days of gray skies, rain and wind.

It was clear that restoring the region to its ordinarily frenetic pace could take days ? and that rebuilding the hardest-hit communities and the transportation networks that link them together could take considerably longer.

"We will get through the days ahead by doing what we always do in tough times ? by standing together, shoulder to shoulder, ready to help a neighbor, comfort a stranger and get the city we love back on its feet," New York Mayor Michael Bloomberg said.

The scale of the challenge could be seen across the Hudson River in New Jersey, where National Guard troops arrived in the heavily flooded city of Hoboken to help evacuate thousands still stuck in their homes. And new problems arose when firefighters were unable to reach blazes rekindled by natural gas leaks in the heavily hit shore town of Mantoloking.

As New York began its second day after the megastorm, morning rush-hour traffic was heavy as people started returning to work. There was even a sign of normalcy: commuters waiting at bus stops.

On the Brooklyn Bridge, closed earlier because of high winds, joggers and bikers made their way across the span before sunrise. One cyclist carried a flashlight. Car traffic on the bridge was busy, and slowed as it neared Manhattan.

By late Tuesday, the winds and flooding inflicted by the fast-weakening Sandy had subsided, leaving at least 55 people dead along the Atlantic Coast and splintering beachfront homes and boardwalks from the mid-Atlantic states to southern New England.

The storm later moved across Pennsylvania on a predicted path toward New York State and Canada.

At the height of the disaster, more than 8.2 million lost electricity ? some as far away as Michigan. Nearly a quarter of those without power were in New York, where lower Manhattan's usually bright lights remained dark for a second night.

But, amid the despair, talk of recovery was already beginning.

"It's heartbreaking after being here 37 years," Barry Prezioso of Point Pleasant, N.J., said as he returned to his house in the beachfront community to survey the damage. "You see your home demolished like this, it's tough. But nobody got hurt and the upstairs is still livable, so we can still live upstairs and clean this out. I'm sure there's people that had worse. I feel kind of lucky."

Much of the initial recovery efforts focused on New York City, the region's economic heart. Bloomberg said it could take four or five days before the subway, which suffered the worst damage in its 108-year history, is running again. All 10 of the tunnels that carry commuters under the East River were flooded. But high water prevented inspectors from immediately assessing damage to key equipment, raising the possibility that the nation's largest city could endure an extended shutdown of the system that 5 million people count on to get to work and school each day. The chairman of the state agency that runs the subway, Joseph Lhota, said service might have to resume piecemeal, and experts said the cost of the repairs could be staggering.

Power company Consolidated Edison said it would be four days before the last of the 337,000 customers in Manhattan and Brooklyn who lost power have electricity again and it could take a week to restore outages in the Bronx, Queens, Staten Island and Westchester County. Floodwater led to explosions that disabled a power substation Monday night, contributing to the outages.

Surveying the widespread damage, it was clear much of the recovery and rebuilding will take far longer.

When New Jersey Gov. Chris Christie stopped in Belmar, N.J., during a tour of the devastation, one woman wept openly and 42-year-old Walter Patrickis told him, "Governor, I lost everything."

Christie, who called the shore damage "unthinkable," said a full recovery would take months, at least, and it would likely be a week or more before power is restored to everyone who lost it.

"Now we've got a big task ahead of us that we have to do together. This is the kind of thing New Jerseyans are built for," he said. President Barack Obama is scheduled to visit the state Wednesday to inspect the storm damage.

By sundown Tuesday, however, announcements from officials and scenes on the streets signaled that New York and nearby towns were edging toward a semblance of routine.

First came the reopening of highways in Connecticut and bridges across the Hudson and East rivers, although the Brooklyn-Battery Tunnel, connecting Brooklyn to Manhattan, and the Holland Tunnel, between New York and New Jersey, remained closed.

A limited number of the white and blue buses that crisscross New York's grid returned Tuesday evening to Broadway and other thoroughfares on a reduced schedule ? but free of charge. Gov. Andrew Cuomo said he hoped there would be full service by Wednesday. Still, school was canceled for a third straight day Wednesday in the city, where many students rely on buses and subways to reach classrooms.

In one bit of good news, John F. Kennedy and Newark Liberty International airports reopened with limited service just after 7 a.m. Wednesday. New York's LaGuardia Airport remains closed.

The New York Stock Exchange was again silent Tuesday ? the first weather-related, two-day closure since the 19th century ? but trading was scheduled to resume Wednesday morning with Bloomberg ringing the opening bell.

Amtrak also laid out plans to resume some runs in the Northeast on Wednesday, with modified service between Newark, N.J., and points south. That includes restoring Virginia service to Lynchburg, Richmond and Newport News, Keystone trains in Pennsylvania, and Downeaster service between Boston and Portland, Maine.

But flooding continues to prevent service to and from New York's Penn Station. Amtrak said the amount of water in train tunnels under the Hudson and East rivers is unprecedented. There will be no Northeast Regional service between New York and Boston and no Acela Express service for the entire length of the Northeast Corridor. No date has been set for when it might resume.

But even with the return of some transportation and plans to reopen schools and businesses, the damage and pain inflicted by Sandy continued to unfold, confirming the challenge posed by rebuilding.

In New Jersey, amusement rides that once crowned a pier in Seaside Heights were dumped into the ocean, some homes were smashed, and others were partially buried in sand.

National Guard troops arrived in Hoboken on Tuesday night to find live wires dangling in the floodwaters that Mayor Dawn Zimmer said were rapidly mixing with sewage.

About 2.1 million homes and businesses remained without power across the state late Tuesday. When Tropical Storm Irene struck last year, it took more than a week to restore power everywhere. The state's largest utility, PSE&G, said it was trying to dry out substations it had to shut down.

Outages in the state's two largest cities, Newark and Jersey City, left traffic signals dark, resulting in numerous fender-benders at intersections where police were not directing traffic. And in one Jersey City supermarket, there were long lines to get bread and a spot at an outlet to charge cellphones.

Trees and power lines were down in every corner of the state. Schools and state government offices were closed for a second day, and many called off classes for Wednesday, too. The governor said the PATH trains connecting northern New Jersey with Manhattan would be out of service for at least seven to 10 days because of flooding. All the New Jersey Transit rail lines were damaged, he said, and it was not clear when the rail lines would be able to open.

In Connecticut, some residents of Fairfield returned home in kayaks and canoes to inspect widespread damage left by retreating floodwaters that kept other homeowners at bay.

"The uncertainty is the worst," said Jessica Levitt, who was told it could be a week before she can enter her house. "Even if we had damage, you just want to be able to do something. We can't even get started."

The storm caused irreparable damage to homes in East Haven, Milford and other shore towns. Still, many were grateful the storm did not deliver a bigger blow, considering the havoc wrought in New York City and New Jersey.

"I feel like we are blessed," said Bertha Weismann, whose garage was flooded in Bridgeport. "It could have been worse."

And in New York, residents of the flooded beachfront neighborhood of Breezy Point in returned home to find fire had taken everything the water had not. A huge blaze destroyed perhaps 100 homes in the close-knit community where many had stayed behind despite being told to evacuate.

John Frawley, 57, acknowledged the mistake. Frawley, who lived about five houses from the fire's edge, said he spent the night terrified "not knowing if the fire was going to jump the boulevard and come up to my house."

"I stayed up all night," he said. "The screams. The fire. It was horrifying."

There were still only hints of the economic impact of the storm.

Forecasting firm IHS Global Insight predicted it will end up causing about $20 billion in damage and $10 billion to $30 billion in lost business. Another firm, AIR Worldwide, estimated losses up to $15 billion ? big numbers probably offset by reconstruction and repairs that will contribute to longer-term growth.

"The biggest problem is not the first few days but the coming months," said Alan Rubin, an expert in natural disaster recovery.

Some of those who lost homes and businesses to Sandy were promising to return and rebuild, but many sounded chastened by their encounter with nature's fury. They included Tom Shalvey of Warwick, R.I., whose 500-square-foot cottage on the beach in South Kingstown was washed away by raging surf, leaving a utility pipe as the only marker of where it once sat.

"We love the beach. We had many great times here," Shalvey said. "We will be back. But it will not be on the front row."

Ken Chelcun of Huntersville has been living without a stomach since Dec. 5, 2008, when he had surgery to remove the digestive powerhouse ? and prevent the cancer that killed his mother and brother.

He?s not the only one in his family who has taken this drastic action.

His sister, nephew and niece have also had their healthy stomachs removed.

All chose surgery after learning they share a genetic mutation that leads to hereditary diffuse gastric cancer, a syndrome that means they have an 80 percent chance of developing stomach cancer.

I wrote about the family in 2008, the same year they started the Chelcun Family Fund to raise awareness and money for research to improve diagnostic testing and treatment for stomach cancer. It has expanded into a worldwide foundation ? No Stomach For Cancer.

On Saturday, Ken Chelcun, his wife, Suzie, and their three sons will participate in the first No Stomach For Cancer walk, around the perimeter of Pine Lake Preparatory school in Mooresville.

Other walks will take place the same day around the world.

Stomach cancer is ?more prevalent than people realize,? Suzie Chelcun said. ?It is the second leading cause of cancer death (worldwide) but receives the least amount in federal funding?. In our experience, it was difficult to find answers. There really wasn?t a place to go.?

The Chelcuns? mother died of stomach cancer in 1982, at age 52. Brother Greg, of Wisconsin, died in 2009, at 58.

After Greg?s diagnosis, Ken Chelcun?s sister, Karen Chelcun Schreiber of Wisconsin, began researching the possibility of an inherited risk for stomach cancer in their family. Blood tests showed that she, Greg and Ken all inherited the genetic mutation.

There is no screening to detect stomach cancer, and symptoms don?t develop until the cancer is advanced.

Given the family history, Ken Chelcun said the choice to have surgery was ?pretty much a no-brainer.?

Now, the next generation is deciding.

Greg?s son Brian, 30, and daughter Johanna, 28, both have tested positive for the mutation and had the surgery.

Schreiber has two grown sons ? one tested positive but hasn?t had surgery; the other has chosen not to get tested.

Ken and Suzie Chelcun?s sons are 14, 12, and 9, and geneticists have advised them to wait until they?re 18 to even consider testing.

?It?s a hard knowledge to face,? Suzie Chelcun said. ?It?s going to be something they?ll have to decide when they get older.?

Toyota Unleashes Its "OctoberBeast" at Annual Company Auto Show

Among car enthusiasts, sometimes you hear jokes like, ?With as boring as the Camry and Corolla are, you wonder what the people who work for Toyota are like.?

The fact is, they?re nothing like you imagine them to be. Many of them are as enthusiastic about cars as your average?Motor Trend?or?Automobile reader. Maybe more so. And they can prove it, too.

OctoberBeast 2 300x187 imageYou don?t typically hear of big, conservative Japanese car companies starting classic car shows in their parking lots, which makes this story all the more interesting. Back in 1992, engineer?Greg Busenkell was searching for a lot around Toyota headquarters in Torrance, California, to put a fleet of cars for new accessories testing. He noticed interesting and classic cars showing up from time to time and figured he might as well try to have them show up at the same time?an annual cruise-in.

Thus, OctoberBeast was born. Or, at least, the Toyota Associate Special Interest Vehicle Display, which is what you probably thought a company like Toyota would call it. But Busenkell quickly renamed it after seeing so many old Detroit ?beasts? show up every October.

The first year found eight cars lined up in a parking lot. Some were show-ready; others were works in progress that would come back year after year with signs of improvement. All of them were loved by their owners, proud of their classics regardless of what badge adorned their hood or grille.

OctoberBeast 31 300x187 imageOctoberBeast continued expanding through the years to include invitations to Toyota?s local suppliers?some years getting as many as 100 cars. While there are no specific rules to which cars are allowed to show up at OctoberBeast, general guidelines are that the vehicle should be at least 25 years old or of special interest. Everything is welcome, whether it runs or not, as OctoberBeast?s official motto has become ?Drive it, Tow it, Push it?Show it!?

Earlier this month, OctoberBeast had approximately 65 cars enter through Toyota?s parking lot, with no classes, awards, or particular arrangement of cars. For Toyota, it has become a social gathering of enthusiastic coworkers who might not otherwise mingle with one another in Toyota?s vast office space. They share their stories and just catch up with one another for the first time in a few months.

OctoberBeast 29 300x187 imageLawrence Keller of Toyota accessories and the torchbearer for OctoberBeast for the last five years, has said during the past 20 years, cars, trucks, dragsters, speedboats, antique strollers have been shown at OctoberBeast. One person even brought a 1940 Piper Cub airplane, which he had to disassemble, load into a truck, and reassemble on the show field. Some of the more memorable cars have been the two ultra-rare Toyota 2000GT sports cars from the company?s private collection, a Porsche Carrera GT, a ?32 Ford hot rod with a Lexus V-8 shoehorned into it, and a pair of original AC Shelby Cobras?289- and 427-cubic-inch engines, respectively.

Says Keller of OctoberBeast, ?This is not a one-man show, and this yearly event could not happen without the assistance and coordination of various departments within Toyota.?

It's Weird, Candidate Height Matters in Elections | Psychology Today

When a 5'9" political science professor (that would be me) talks with his 6'7" co-author (that would be Dave Schmitz), it is not surprising that the conversation can turn to the ?weird? fact that the taller candidate has won the popular vote in 67% of all U.S. presidential elections. While interesting water cooler conversation and often a tidbit dispensed by prognosticating political pundits, this oddity seems to be more than a statistical fluke and worthy of some explanation.?

But first, how do our current contenders measure up? Barack Obama is 6'1", and challenger Mitt Romney is 6?2?. Our argument, which is grounded in evolutionary theory, does not say that Romney will win because he is the tallest candidate. It suggests that we voters prefer our leaders in general to be tall. And the fact that both of these candidates are taller than the average U.S. male citizen, who is my size at 5'9", is completely consistent with this argument. Also consistent with this argument is the fact that Romney?s major Republican challengers Rick Perry, Newt Gingrich, Herman Cain, and Ron Paul each are 6'0" or taller.?

So why would we prefer taller leaders? Our explanation relies on evolutionary psychology, which suggests that human behavior is the result of not only people's environment, such as things they learn from their parents, but also long-term evolutionary forces, such as psychological mechanisms that evolved to solve our ancestors' problems regarding survival and reproduction. Although it may seem irrational in the modern context, we may prefer taller leaders because our ancestors who selected more formidable allies (height is a common cue for physical formidability) were more likely to survive and pass on related genes in their violent environment. In other words, when our ancestors had a big, strong friend who would help them acquire and protect resources (e.g., food, shelter, and territory), they had a relative advantage in terms of living longer and producing more children.?

Studies of all kinds of animals show that greater physical size and higher social rank go together. This relationship has been found in a wide range of nonhuman animals, from our closest relatives, chimpanzees, gorillas, and baboons, to African elephants, Red Deer, and even to varieties of birds and fish.?

In the realm of human animals, anthropologists measuring skeletons in pre-classical Greek and ancient Mayan graves found a clear association between greater physical stature and "political control" (those anthropologists have all the fun). Further, modern humans tend to perceive individuals with greater authority status as taller than they actually are and, conversely, to perceive taller individuals as having higher professional status. (This study was based on rating professors, and yes, as an average-height professor, this worries me a little). Research in a variety of countries also shows, among other advantages, that taller males make more money and get more job promotions. And finally, in politics specifically, winners of elections are perceived as being taller after than before the election, while losers suffer yet another indignity and are perceived as shorter.?

With this background research in mind, to test our argument about a preference for leaders with greater physical stature we conducted a study that took the perspective of followers (versus leaders). We gave our subjects, U.S. and international undergraduate students at different schools in different parts of the country, three tasks: draw the "ideal national leader," draw "the typical citizen," and, finally, draw a meeting of the leader and citizen. Because we expect people to prefer more physically formidable leaders, we predicted our subjects would tend to draw a taller leader meeting a shorter citizen, with height measured by the vertical size of the figures.?

In fact, that is what we found. More than twice as many subjects (64%) drew a taller leader than drew a taller citizen (31%). And remember the reference to the international students? We found similar patterns in their responses, which suggests the preference for leaders with greater physical stature not only exists in the U.S. but also in different cultures around the world. This is important evidence to evolutionary psychologists, because they argue that a universal behavior cannot be explained by people learning from their specific environment; therefore, evolutionary forces, not environmental-cultural forces, are the most likely explanation for the behavior.?

So we know this election?s crop of presidential candidates measures up, at least in these evolutionary terms, but let?s hope they also measure up when it comes to leading the country.?

It's taken its sweet time showing itself but we've finally got to handle Samsung's first Windows Phone 8 device, the ATIV S (not to be confused with the similarly-named Windows RT tablet). At Microsoft's launch event for its new mobile OS, the phone's 4.8-inch AMOLED display certainly helps to make the most of those colorful tiles -- this time around a higher resolution (1,280 x 720) screen kept everything looking sharp. Aside from the screen, the hardware doesn't pack a unibody build, but it does offer the power user both a 2,300 mAh battery and the microSD slot -- a combination that's looking pretty rare among debut Windows Phone 8 devices. On the other hand, Samsung's brought its (admittedly divisive) glossy plastic shell of its Android Galaxy phones, although there is some cosmetic differences. We take a tour of the hardware -- and that freshly minted OS -- after the break.

Hulk Hogan settles sex tape lawsuit with DJ

CLEARWATER, Fla. (AP) ? Former pro wrestler Hulk Hogan has settled a lawsuit with a disc jockey over a sex tape involving Hogan and the DJ's wife.

A statement by Hogan's publicist Monday says the terms of the settlement are confidential.

Hogan said in a lawsuit earlier this month that he had consensual sex with his best friend's wife, Heather Clem, six years ago in the Clems' home, but he didn't know he was being videotaped.

The disc jockey, Bubba the Love Sponge Clem, apologized to Hogan on Monday. Clem also said that he's convinced Hogan was unaware he was being filmed.

A lawsuit is still pending against the gossip website Gawker, which put excerpts of the video on its site.

Delta Epsilon Mu Looks to Join UA | The Arkansas Traveler

Fraternity members from Delta Epsilon Mu are looking to start a chapter at the UA.

Delta Epsilon Mu is a national professional fraternity geared toward students interested in careers in the health, medicine, physical therapy, dentistry, nursing or biomedical engineering fields.

?I think they?re just looking to expand,? said Shane Barker, pre-professional advisor for Fulbright College. ?It?s very preliminary, and I?m trying to help facilitate getting them here on campus.?

A handful of students at the UA are interested in being a part of the founding chapter of DEM, said Michael Dawar, director of expansion and president of the DEM chapter at the University of Kansas.

?I was contacted by students at the university over the summer stating they were interested in starting a chapter of our fraternity at their university,? Dawar said. ?They requested assistance in recruiting members, so I started posting on Facebook pages and groups to gain attention for the group and help them recruit more students.?

The UA would be one of the few DEM fraternities in the Midwest, which would allow students to work with other nearby chapters and get involved on a national level, Dawar said.

?Our fraternity was interested in starting a chapter of our fraternity at Arkansas because we were told there was no other organization of our kind at the school, and that it would help unite students of various health fields under one banner,? Dawar said.

Dawar said he hopes a chapter can be established at the UA by the end of this semester or the beginning of the spring semester.

There are already four registered student organizations on campus for pre-dentistry, pre-medicine, pre-optometry and pre-pharmacy students, but Barker said there were no organizations for students in other popular medical fields such as physical therapy.

?I was looking to do something for them either individually or organizationally,? Barker said. ?It was perfect timing both ways.?

All students are welcome to join DEM even if they are already involved with a pre-heath RSO, Barker said.

?Because we are pre-health, we cover all the different health fields, so members have an opportunity to learn about all the fields and see what will best fit their interests,? Dawar said.

DEM offers professional and personal development workshops, speakers from the health field, community service and volunteer opportunities and extensive networking opportunities across the nation, Dawar said.

?Many of our members are in similar majors and career paths, so they are able to study together and help the younger members with classes they have already taken,? Dawar said.

They also help members with job shadowing, preparing for graduate schools in their field, getting into graduate programs, getting into contact with admissions representatives from schools and visiting schools as a group.

DEM is the only national professional fraternity that is open to both men and women, according to the DEM website.

DEM has 12 chapters and seven colonies on 14 university campuses throughout the country, and it is the fastest growing pre-health professional fraternity in the nation, according to the DEM website.

China's leadership shakeup: Am I an unfortunate casualty?

I've been trying ? and trying ? to reinstall software I need to freely access the Web in China. I increasingly suspect I'm doing battle with state-sponsored hackers ahead of the sensitive party congress.

I have just spent an entire day wrestling with my computer and my Internet connection, and I have a strong suspicion that I have been wrestling too, at a distance, with an agent of the Chinese government who has been doing his or her best to frustrate me.

In order to access the Web freely from China, you need what is called a Virtual Private Network, which jumps the Great Firewall erected by Chinese censors. Mine expired the other day, so I needed to re-install it.?

That proved unusually difficult, even with online help from the company selling me the VPN, and it became clear that something was just not right.

My suspicions were heightened by the fact that I, like many other journalists, have recently received emails with Trojan horse malware (malicious code that looks like a legitimate file but in fact gives a hacker access to a computer) in their attachments. Cyber analysts who inspected them have warned that the attachments appear to come from state-sponsored hackers.?

Google has also informed me in a banner appearing on my Gmail account that it suspects "state-sponsored" hackers have been trying to penetrate my account.?

The last time this happened to me was during the Tibetan riots in 2008, when the authorities were very, very nervous about foreign journalists and began interfering directly with our communications. (That is over and above the normal surveillance to which our emails and phone calls are subject.)

Today we are at another highly sensitive political juncture, 10 days away from the 18th?congress of the ruling Communist Party, which is due to anoint a new generation of leaders. But there are signs of a continuing power struggle at the very top of the party, suggesting that the government system is a good deal less stable than Beijing would like us to believe.

There came a moment this afternoon, when the VPN would not install, when a Microsoft update would not install, and when a virus detector would not install, that I came to believe I was in direct contact with my persecutor.

I was on the ?Sophos? virus detector?s webpage, seeking to download the tool. Each time I clicked on ?download,? I got the standard message when the censors have banned a site: ?Internet Explorer cannot display the webpage.? But the page itself was not blocked and after a few tries I found I was being cut off even before my cursor reached the ?download? button.

It was just as if somebody was watching my screen and interrupting me as I was on the point of doing what I wanted to.

I have no idea how possible this is, but tend to take the advice of my Chinese assistant. ?There is nothing a hacker cannot do,? she has decided. ?Why don?t you try again when ?they? have gone off duty??

So I?ll be back in the office at midnight, and hope that ?they? do not work 24/7?

Cyprus police head off hit on country's top lawyer

(AP) ? Cyprus police say they have thwarted an attempt on the life of Attorney-General Petros Clerides.

A police official speaking on customary condition of anonymity says three Cypriot suspects now in police custody face a charge of conspiracy to commit murder.

The motives for the alleged murder attempt are unclear.

The official says that police made the arrests after one of the suspects received an arms cache that included an anti-tank missile launcher.

The suspects will appear in a Nicosia court later Monday for a hearing to determine whether they should remain in police custody until investigators are ready to formally charge them.

?Without a Fight at UNC's Memorial Hall | Triangle Arts and ...

Durham?s Megafaun to perform live score of acclaimed documentary, Without a Fight, at UNC?s Memorial Hall

UNC?s Center for Global Initiatives, Carolina for Kibera will host a one-time only screening of the acclaimed documentary Without a Fight, featuring a live musical accompaniment from the Durham-based band Megafaun and a host of special guests at UNC?s Memorial Hall Thursday November 1 at 7 pm.

Without a Fight follows the triumphs and travails of four African youth as they vie for a championship in Kibera, a Kenyan slum rife with ethnic tensions and violent upheavals. For the youth of Kibera, soccer is a way of life. But until the Champion?s League was formed ten years ago, thanks in part to the UNC-based nonprofit Carolina for Kibera, soccer was also a source of tension and violence. Youth peace activists in Kibera asked themselves a question in the face of tribal tension: what would happen if they formed a league that required kids from different tribes to be on the same team? Without a Fight answers this question and offers a unique look into the problems of poverty and division.

The world premier of Without a Fight was held at 11 mm Film Festival in Berlin in March, and the North American premiere was held at Full Frame Documentary Film Festival in April. Since then, the film has been shown at universities across the country. Without a Fight has been nominated and short listed for a special jury award by the Peace & Sports International Forum. In mid November, the film will screen as an official entry at the All Sports LA Film Festival

The screening at Memorial Hall marks a homecoming for director Jason Arthurs, who graduated from Carolina, and producer Beth Kutchma, who works at UNC?s Center for Global Initiatives and is part of the heralded Durham-based band Red Collar. While finishing Without a Fight, they asked Megafaun?s Brad and Phil Cook to write and compose the soundtrack. They did, and at Memorial Hall November 1 the Cook brothers will perform a live rendition of the score they composed.

At 7pm, UNC alum and Carolina for Kibera co-founder Rye Barcott will introduce the trailer to his book, It Happened on the Way to War, which inspired Arthurs and Kutchma to make Without a Fight. Acclaimed singer-songwriter JKutchma will perform his song Arms Around the World, which he composed for Barcott?s trailer.

After the screening of Without A Fight, there will be a Q&A session with Barcott, Kutchma, Arthurs, Phil and Brad Cook, and Hillary Omala, the Executive Director of Carolina for Kibera?the organization that sponsors the soccer league featured in the film.

Tickets to the screening are $15 for the public, $10 for UNC faculty and staff, and $5 for students with ID. Children under 12 are free.

Novel genes that may drive rare, aggressive form of uterine cancer identified

ScienceDaily (Oct. 28, 2012) ? Researchers have identified several genes that are linked to one of the most lethal forms of uterine cancer, serous endometrial cancer. The researchers describe how three of the genes found in the study are frequently altered in the disease, suggesting that the genes drive the development of tumors. The findings appear in the Oct. 28, 2012, advance online issue of Nature Genetics. The team was led by researchers from the National Human Genome Research Institute (NHGRI), part of the National Institutes of Health.

Cancer of the uterine lining, or endometrium, is the most commonly diagnosed gynecological malignancy in the United States. Also called endometrial cancer, it is diagnosed in about 47,000 American women and leads to about 8,000 deaths each year.

Each of its three major subtypes -- endometrioid, serous and clear-cell -- is caused by a different constellation of genetic alterations and has a different prognosis. Endometrioid tumors make up about 80 percent of diagnosed tumors. Surgery often is a complete cure for women with the endometrioid subtype, since doctors usually diagnose these cases at an early stage.

Compared to other subtypes, the 2 to 10 percent of uterine cancers that comprise the serous subtype do not respond well to therapies. The five-year survival rate for serous endometrial cancer is 45 percent, compared to 65 percent for clear-cell and 91 percent for endometrioid subtypes. Serous and clear-cell endometrial tumor subtypes are clinically aggressive and quickly advance beyond the uterus.

"Serous endometrial tumors can account for as much as 39 percent of deaths from endometrial cancer," said Daphne W. Bell, Ph.D., an NHGRI investigator and the paper's senior author. Dr. Bell heads the Reproductive Cancer Genetics Section of NHGRI's Cancer Genetics Branch.

To determine which genes are altered in serous endometrial cancer, Dr. Bell and her team undertook a comprehensive genomic study of tumors by sequencing their exomes, the critical 1 to 2 percent of the genome that codes for proteins.

"Exome sequencing is a powerful tool for revealing important insights about this form of cancer that exacts such a high toll for thousands of women," said NHGRI Scientific Director Dan Kastner, M.D., Ph.D. "This study pinpoints genetic alterations that may be essential for onset and progression of uterine cancers and may eventually lead to new therapeutic targets."

Dr. Bell's team focused on the rarer, more aggressive forms of endometrial cancer. They began their study by examining serous tumor tissue and matched normal tissue from 13 patients. National Cancer Institute and Massachusetts General Hospital pathologists processed the 26 tissue samples, which subsequently underwent whole-exome sequencing at the NIH Intramural Sequencing Center.

With the exome data in hand, the researchers filtered through millions of data points to locate alterations, or mutations. They disqualified from the analysis any mutation found in a tumor and its matched healthy tissue, looking expressly for mutations that occurred exclusively in the tumor cells. They also eliminated one of the 13 tumors from analysis because its exome had hundreds more unique mutations than any other tumor.

The researchers detected more than 500 somatic mutations within the remaining 12 tumors. They next looked for genes that were mutated in more than one of the tumors. An alteration that occurs in more than one tumor is more likely to be relevant to the development of the cancer than a unique alteration.

"When you identify a set of mutations, they could either be drivers that have caused the cancer or incidental passengers that are of no consequence; our goal is to identify the drivers," Dr. Bell explained. "One way to do this is to home in on genes that are mutated in more than one tumor, because we know from experience that frequently mutated genes are often driver genes."

The team felt confident that alterations in nine genes could be driver genes in serous endometrial cancer. Three of the nine genes had previously been recognized by researchers in the cancer genetics field as a cause of serous endometrial cancer. To get a clearer picture of driver gene status among the other six genes, the researchers sequenced each gene in 40 additional serous endometrial tumors. They discovered that three of the six genes -- CHD4, FBXW7 and SPOP -- are altered at a statistically high frequency in serous endometrial cancer.

The team also found that this set of three genes is mutated in 40 percent of the serous endometrial cancer tumors and in 15 to 26 percent of the other endometrial cancer subtypes.

Probing still further, the researchers looked for the same genes highlighted by their exome sequencing study within databases that organize genes according to their biological function. They found an enrichment of genes involved in chromatin remodeling, the dynamic process by which the contents of the cell nucleus, including DNA, are packaged and modified. Chromatin remodeling enables tightly packaged DNA to be accessed for the expression of genes. Intriguingly, CHD4 was one of the genes that formed the chromatin-remodeling cluster.

"We sequenced the other genes that make up this cluster and, as a set, these genes are frequently mutated in both serous and clear-cell endometrial tumors," said Dr. Bell.

They also noted frequent mutations in genes that regulate a process known as ubiquitin-mediated protein degradation. The process targets unneeded proteins for destruction, and thus prevents them from accumulating within the cell. Left to accumulate, some of the target proteins are known to drive cancer formation. FBXW7 and SPOP are both known to play a role in binding to the unneeded proteins and targeting them for destruction.

Many of the FBXW7 gene mutations that Dr. Bell's team identified are known in other cancers to be driver mutations that prevent the FBXW7 protein from binding to its target protein. Dr. Bell believes that altered SPOP may behave the same way. "All the mutations we found in SPOP are in the region that binds the target proteins" she said. "We suspect the mutations in SPOP might lead to the accumulation of the unneeded proteins within the cell. But that has to be tested."

The current findings build on the team's 2011 study that showed for the first time that alterations in the PIK3R1 gene occur in all subtypes of endometrial cancer and are most frequent in the more common endometrioid subtype.

"This discovery really changes our understanding of some of the genetic alterations that may contribute to this disease," Dr. Bell said, acknowledging that the findings are limited by the small number of tumors subjected to exome sequencing.

She noted that it is too early to make a direct connection between their findings and prospects for treatments for this aggressive form of uterine cancer.
